What follows is my thoughts and speculation about the spoilers of this week's episode from the Space Channel Promo.

And it is written that the first seal shall be broken
when a righteous man spills blood in Hell.
This is the big reveal Alastair gives to Dean. It's a savage blow and there's no wonder that it shatters Dean. First impression....stunned. But it works so well. The spoiler, misleading as they sometimes are, stated that it was Dean being pulled from Hell that caused the seals to be broken. This never did make much sense, so I'm glad it was wrong. Not much else could be worse for Dean, to learn that he is the reason for the coming apocalypse. I can only imagine how much more reckless this might make him. Keeping the seals from being broken will turn into something much more personal. I hope Sam learns of this mainly because the boys don't need anymore lies between them.
And Dean...poor, bearing the weight of the very world on his shoulders Dean. He held out for 30 years of being tortured in Hell. He's always had self-esteem issues and this will only make them worse. If only he'd held out for another 10 years this never would have happened. It's a devastating blow to an already fragile soul. I can only hope that finds a way out of the hole of despair he's sure to fall into at this news....and that Sam is there to help him climb back out.
I do have 2 issues with this statement about Dean being the reason the seals start to get broken:
1) Righteous - Defined as "acting in accord with divine or moral law : free from guilt or sin" by the Merriam-Webster Dictionary. I would hardly think Dean is free of guilt or sin, but maybe the rules in Hell are more lenient?
2) All these thousands of years have gone by without the seals being broken. You're telling me there hasn't been a righteous man to spill blood in Hell all this time?
Now let's go to the second big reveal of the Space Promo. Sam is apparently drinking Ruby's blood. I'm assuming this is to bulk up his demon extraction powers. Spoilers for upcoming episodes indicate that Sam may well be addicted to the demon blood. As for the blood sucking, I'm sure everyone's inclination is to think that he's now going Dark Side. He may well be, but here' where my theory comes in.
I'm speculating that Sam thinks he can control his powers. He told Dean as much at the beginning of this season's "Metamorphosis" ("I won't let it go that far."). Sam wasn't able to keep Dean from going to Hell. He even failed to get Dean out of the pit. Those are two heavy weights of guilt on Sam and the only way he can get any satisfaction, the only way he figures he can help Dean now, is to get revenge on Lilith for sending Dean to Hell in the first place. Going after Lilith is Sam's singular goal right now. If that means losing himself to his demonic powers for a while, so be it. He's probably convinced himself that once Lilith is dead he can give up the demonic blood addiction along with the powers. And of course, he's not going to let his brother know what he's been up to - hanging out with Ruby and building his powers - because Dean would naturally try to stop him. So the way I see it, Sam's intentions are good (or he at least believes they're good). He's trying to protect Dean, to keep his brother alive. Because after seeing Sam during those 4 months of Dean being gone, I'm sure he wouldn't be able to take that loss again.
My guess about 4x16 is that Sam, worried about Dean losing himself to his own darkness in torturing Alastair, goes to Ruby to get a demon blood pick-me-up so he can take out Alastair. The episode stills seem to back up this theory.
